Description:

The mutations induced in approximately 2,000 mutants of Salmonella by the heterocyclic@ amine Glu-P-1 were determined by colony probe hybridization and PCR/DNA sequence analysis. ll of the mutations were at sites containing guanine, which is the base at which Glu-P-1 forms DNA adducts. t the frameshift allele, Glu-P-1 induced essentially 100% GC or CG deletions within the sequence CGCGCGCG. he DNA repair background did not influence this spectrum of mutations. t a base-substitution allele, Glu-P-1 induced 75% GC to TA transversions and 25% GC to AT transitions all at a single site (the middle C of a CCC codon). his distinct site specificity as well as Glu-P-l's limited (20-30 times less) base-substitution mutagenic potency relative to its frameshift potency may have bearing on the lack of base substitutions identified in ras genes in Glu-P-1-induced rat colon tumors.
