Perchlorate exposure and association with iron homeostasis and other biological functions among NHANES 2005-2008 subjects Schreinemachers DM, Ghio AJ, Cascio WE, Sobus JR. U.S. EPA, RTP, NC, USA Perchlorate (ClO4-), an environmental pollutant, is a known thyroid toxicant and oxidant. Its association with thyroid hormone levels has been presented in several studies. Associations with other biological measures that may or may not be dependent on thyroid hormone signaling, are largely unknown. This study investigates the associations between perchlorate exposure and serum iron homeostasis, antioxidants, renal function, lipids, and blood cell counts among NHANES 2005-2008 subjects. Most analyses were performed on healthy males (n=3705) and non-pregnant healthy females (n=2967), ages 12-59, that had not been diagnosed previously with cancer, heart failure, coronary heart disease, angina, heart attack, stroke, thyroid problems, diabetes, or prediabetes, and were not receiving antineoplastic, cardiovascular, hormonal, or metabolic prescribed drugs in the month prior to the interview. Linear regression models controlling for potential confounders, identified significant associations of creatinine-adjusted perchlorate with serum uric acid, serum iron, blood urea nitrogen, red cell and lymphocyte count for both males and females. These results are presented as the change of the natural log-transformed biomarker level per unit increase of log-transformed, creatinine-adjusted perchlorate (β, 95% CI): serum uric acid, males -0.033, (-0.044, -0.023), females -0.027 (-0.038, -0.016); serum iron, males -0.064 (-0.085, -0.043), females -0.053 (-0.081, -0.025); RBC count, males -0.008 (-0.013, -0.004), females -0.006 (-0.010, -0.002); blood urea nitrogen, males 0.029 (0.011, 0.046), females 0.045 (0.025, 0.065); lymphocyte number, males 0.019 (0.004, 0.034), females 0.030 (0.011, 0.048). The results indicate that exposure to perchlorate may be associated with measurable changes in biochemical outcomes suggestive of oxidant stress and altered serum iron regulation.
