Numerous surface waters are adversely impacted by contaminants released from sources such as WWfPs, CAFOs, mining activities, and agricultural operations. Ideally, an assessment strategy for these applications would include both chemical identification and effects-based monitoring. However, traditional effects testing methods are resource and time intensive and may not be sustainable in the long term, given the large number of ecosystems that require monitoring. Moreover, conventional effects testing often utilize large numbers of animals, making its continued use unsustainable give? existing efforts to reduced animal use. Finally, only a handful of endpoints are typically measured using conventional approaches, limiting the availability of potentially critical toxicity information. A promising approach for addressing all of these concerns is cell culture-based metabolomics. By combining optimized cell-culturing techniques with NMR-based metabolomics, we have developed a relatively low cost, high throughput approach for obtaining multi-endpoint, molecular level responses using ambient water from environmentally impacted sites. Here we will describe the results of applying these techniques to assess the impacts of field-collected water samples from contaminated and reference sites on biological processes of Zebrafish (Danio rrrio) liver cells (ZFL). These data will be interpreted in the context of an extensive set of site and chemical characterization data that will also he measured on these waters.
