Potency determination is important to identify the most promising drug candidates as well as identification of and ranking of compound toxicity. In our laboratory, we have utilized MEA recording techniques to determine the relative potency of 11 insecticidal compounds and rank their relative potency. Primary cultures of rat cortical neurons were grown on glass MEA electrodes for a period of 9-16 days, when steady state spontaneous spiking activity was reached. Cumulative concentration-response determinations of the inhibition of spontaneous spiking behavior included assessment of up to 6 concentrations of compound on each MEA. The use of a four amplifier system significantly accelerated this approach. Following determination of the EC50 values for each of 11 different insecticidal compounds, the relative potencies were determined by expressing the potency of each relative to a selected reference compound, and these relative potencies ranged from
