Impact/Purpose:
This presentation was invited for the OECD working group on developmental neurotoxicity. The purpose of the talk is to update the group on the progress of building a predictive model for DNT using the in vitro DNT battery data in ToxCast. The presentation will cover topics such as differences in bioactivity results using different pipelines and the concordance between assays that measure the same neurodevelopmental process.
Description:
The meeting aims to 1) discuss progress towards updating the Initial Recommendations to an OECD guidance, 2) discuss the regulatory implementation of the current DNT-IVB and strategy to include/exclude additional assays, and 3) discuss complementary aspects of the guidance that are relevant to the regulatory use (i.e., tiered testing, (Q)IVIVE, uncertainty comparison table). Updates on Chemicals Tested in the DNT IVB Assays, data availability and update of Appendix E.Sharing methodologies to analyse data and ways to come up with one interpretation.Advances analysis and integration of the existing DNT-IVB data and opportunities to• Understand and explain differences in data from assays that measure the same developmental process.• Understand the sensitivity of some methods in the battery and advise for which purpose they should be used or not.• Use machine learning to solve the problem of integrating data from the battery.Opportunity to share advancements in mathematical models and considerations about
