The purpose of this project is to investigate the estrogen response in zebrafish with particular attention paid to embryos and young fish. One means will be to investigate the expression of the zebrafish estrogen receptor during these times using whole mount in situ hybridization. Some of this will be done with fluorescence so that we can three-dimensionally reconstruct and localize the estrogen receptor mRNA during embryogenesis and to also examine its expression under the influence of embryo treatment with estrogens. A second approach will involve the examination of expression from an introduce estrogen responsive reporter gene that provides a localized fluorescent signal (the green fluorescent protein gene). With this reporter we hope to record gene expression within the whole organism and capture this using the laser scanning confocal microscope.
