The Research Project on Growth and Development (G&D) is designed to achieve Specific Aim 1: to test the hypothesis that prenatal exposure to PAH (a component of particulate matter less than 2.5 microns/ PM2.5) and prenatal and/or postnatal exposure to ETS are significant contributors to impaired fetal and early childhood growth and development, after controlling for the effects of exposure to lead and other possible confounding variables. Further, to test the hypothesis that impaired nutritional status (inadequate levels of antioxidants and essential fatty acids) act as a susceptibility factor to increase the effects of the environmental exposures (PAH and ETS) on these adverse outcomes. The G& D project is a prospective molecular epidemiologic study of 400 infants whose mothers are enrolled before the last trimester of pregnancy. Enrollment procedures will ensure that the sample is evenly distributed across the geographic area in order to capture variations in environmental exposures as well as community-level socioeconomic and other risk factors. The 400 infants will be followed prenatally and through the first 2 years of life. Questionnaires will be administered to the mothers by Research Workers at enrollment, prenatally and at serial timepoints during the first 24 months of life to obtain information on environmental exposures (including smoking, home heating and cooking sources), socioeconomic status (SES) and response to socioeconomic stress, specific food sources, daily activities and health status. Personal exposure to PAH will be monitored prenatally and ambient prenatal and postnatal PAH exposure will be assessed using the GIS. Umbilical cord blood and maternal blood samples will be collected at delivery, as will a 24 month blood sample from the infants. Biomarkers (PAH-DNA adducts, cotinine, lead, antioxidants, essential fatty acids) will be analyzed in those samples. A home environment stimulation inventory will be obtained. Birth outcomes and growth and development during the first 2 years will be evaluated using the Fagan Test and the Denver and Bayley Scales. Relevant data will be shared with both the Asthma Research Project & the Community-Intervention Project. Researchers on the study will maintain close contact with the participants to foster relationships, facilitate compliance and minimize attrition. Supplemental Keywords:
